A brief explanation of this estimation is as follows: Assume that the extinction coefficient is the same for the C=O stretching band arising from each secondary structure. Then the absorption intensity (area of band) is proportional to the number of the C=O groups involved, i.e., the number of the residues. Therefore, the intensity ratio among the bands is proportional to that of the number of residues contained in the particular secondary structure.
