••] show that high expression of PU.1 is non-permissive for T- and B-cell development from fetal progenitors in fetal thymic organ cultures (FTOCs), but can instead be conducive for macrophage development. In normal hematopoietic cell development PU.1 may be required to maintain the myeloid differentiation program and inhibit lymphoid-lineage-specific gene expression.

Transient IL-7/IL-7R signaling provides a mechanism for feedback inhibition of immunoglobulin heavy chain gene rearrangements
-
••] is an impressively executed study on the rhythmic and transient nature of IL-7 signaling, which is essential for the maintenance of accessible state of chromatin necessary for gene rearrangement. Diminished IL-7 signaling at pre-B cells results in hypoacetylated VH genes and termination of further gene rearrangement, perhaps contributing to the establishment of allelic exclusion.

-
-/- mice to date. The catalytic p110δ subunit is preferentially expressed in hematopoietic cells. Although the knockout mice show profound defects in mature B-cell activation, precursor B-cell subsets are present in normal numbers. Interestingly, B1 B cells and marginal zone B cells are lacking in the mutant mice, indicating selective functions of PI3K in distinct B-cell subpopulations.

A role for Wnt signalling in self-renewal of haematopoietic stem cells
-
This paper marks the definitive arrival of the canonical WNT signaling in HSC proliferation. Activation of WNT signaling in HSCs in vitro leads to upregulation of HoxB4 and Notch1 and increased cell cycle progression. Inhibition of WNT signaling results in diminution in proliferation and in vivo reconstitution capability of HSCs.

Sequential roles of Brg, the ATPase subunit of BAF chromatin remodeling complexes, in thymocyte development
-
This work is an impeccable examination of T-cell development in mice lacking Brg
-
Chi T.H., Wan M., Lee P.P., Akashi K., Metzger D., Chambon P., Wilson C.B., Crabtree G.R. Sequential roles of Brg, the ATPase subunit of BAF chromatin remodeling complexes, in thymocyte development. Immunity. 19:2003;169-182 This work is an impeccable examination of T-cell development in mice lacking Brg, an integral component of the mammalian BAF complex. Brg-deficient cells do not differentiate, are susceptible to die (perhaps owing to the loss of Bcl-2 expression) and are cell cycle arrested. Pro-T cells lacking Brg show a selective loss of c-Kit expression as well as expression of other WNT target genes, suggesting that the WNT signaling is defective in Brg-deficient cells.

w/w mice that showed relatively normal T- and B-cell development

Pim serine/threonine kinases regulate the stability of Socs-1 protein
-
Results from this report indicate that IL-7 activation of Pim1 maintains high levels of SOCS1 by phosphorylation. Consistent with the regulatory function of Pim serine/threonine kinase
-
-/- thymocytes exhibit diminished levels of SOCS1 protein.
