The mice were sacrificed at 40 min post-injection and their brains were dissected, snap-frozen in liquid nitrogen, and homogenized, followed by addition of 1 mL of isotonic saline. The mixture was vigorously vortexed and 0.5 mL of MeCN was added. After centrifugation for 5 min at 6.000g the supernatant was collected. Samples from the extract and the tissue were counted using a γ well-counter to determine the extraction efficiency (>95% was extracted). The extract was spiked with authentic 4 and analyzed by reverse-phase HPLC using methods described above for the analysis of radioactivity distribution in plasma
