    • Specific labeling of cell surface proteins with chemically diverse compounds
    • 1 in HEK293 cells were covalently labeled with coenzyme A derivatives bearing cyanine fluorophores or a biotin affinity handle. The dependence of the labeling reaction on enzyme activity can also be exploited for exceptional spatial control of labeling by immobilizing the enzyme via genetic fusion.

Thus, it was demonstrated that Syt I plays a role in synaptic vesicle endocytosis, in addition to its well-documented role in vesicle exocytosis. Direct testing of the in vivo role of Syt I specifically during synaptic vesicle endocytosis was made possible by the combination of site-specific protein labeling and fluorophore-assisted light inactivation.
