Describes a novel methodology for the rapid diagnosis of intracellular flux responses based on metabolic flux ratios from GC-MS-derived mass distributions in proteinogenic amino acids. A first demonstration of the potential for high-throughput is given by the analysis of 68 E. coli shake flask cultures, by far the highest number of analyses reported in a single study to date. Moreover, the technical and biological robustness of the method was demonstrated by investigating the influence of key factors like culture conditions, harvest time, different isotopic tracers and known mutations.
