Hall Cdc42 regulates GSK-3β and adenomatous polyposis coli to control cell polarity Nature 421 2003 753 756 This paper describes a signalling pathway involved in cell polarisation in migrating astrocytes. The small GTPase Cdc42, acting through the complex of Par6 with PKC ζ, induces the inactivation of GSK3β at the leading edge. This allows APC, one of the targets of GSK3β, to bind to the MT plus ends and leads to the polarisation of the MT network.

Pellman Cell cycle control of kinesin-mediated transport of Bik1 (CLIP-170) regulates microtubule stability and dynein activation Dev Cell 6 2004 815 829 The authors demonstrate that in budding yeast the kinesin Kip2p delivers the homologue of CLIP-170, Bik1p, to the MT plus ends. The plus-end-directed migration of Bik1p-Kip1p complexes is shown directly by fluorescent imaging. Bik1p, in its turn, stabilizes MTs and participates, together with dynein, in the positioning of the mitotic spindle. The activity of Bik1p and dynein at the MT tips is thus controlled by the levels of Kip2p, which are elevated during mitosis.

    • K.E. Busch, and D. Brunner The microtubule plus-end-tracking proteins mal3p and tip1p cooperate for cell-end targeting of interphase microtubules Curr Biol 14 2004 548 559 The paper describes the function of the fission yeast homologue of EB1, Mal3p, in organising the MT network. Mal3p is shown to promote the initiation of MT growth and inhibit catastrophes. Mal3p is also needed for the proper localisation of Tip1p to the MT plus ends, possibly because Tip1p and Mal3p interact directly. In addition to accumulating at the growing MT tips, Mal3p is also observed in as-yet-unidentified particles moving to the MT minus end.

    • M. van Breugel, D. Drechsel, and A. Hyman Stu2p, the budding yeast member of the conserved Dis1/XMAP215 family of microtubule-associated proteins is a plus-end-binding microtubule destabilizer J Cell Biol 161 2003 359 369 This in vitro study of the properties of the yeast member of the XMAP215 family, Stu2p, shows that it can recognize and preferentially bind to the plus ends of taxol-stabilized MTs. Stu2p has a destabilizing effect on MTs grown in vitro by decreasing the MT growth rate and increasing the frequency of catastrophes. This result is confirmed in vivo by observation of a temperature-sensitive Stu2 mutant.

Glover Mutations in orbit/mast reveal that the central spindle is comprised of two microtubule populations, those that initiate cleavage and those that propagate furrow ingression J Cell Biol 166 2004 49 60 Imaging experiments in Drosophila spermatocytes demonstrate that during cytokinesis the central spindle is composed of two distinct MT populations: 'peripheral' MTs, which initiate the cleavage furrow, and 'interior' MTs, which promote the furrow ingression. Orbit/MAST strongly accumulates at the interior central spindle and in Orbit/MAST hypomorphic mutants the interior MTs are unstable and fail to recruit Pav-KLP and AuroraB. This leads to defects in the formation of the actin contractile ring and a cytokinesis failure.

Earnshaw Human CLASP1 is an outer kinetochore component that regulates spindle microtubule dynamics Cell 113 2003 891 904 Human CLASP1 is identified as a component of the kinetochore 'outer corona', which is the outermost part of the kinetochore and makes the contact with the MT plus ends. By overexpressing the MT-stabilizing portion of CLASP1 and microinjecting anti-CLASP1 antibodies, the authors show that CLASP1 is needed for maintaining mitotic spindle bipolarity and the normal dynamics of kinetochore-attached MTs. These findings are in line with the previous observations by the same authors in Drosophila cells [47].
