    • Results shown here contradict the previous interpretation of Xu et al. (4), who suggested the persistence of cyclical KaiC induction even in the dark. However, according to recent retrospective comments by the above authors (6), nonphosphorylated KaiC in their Western blots (4) was obscured by high nonspecific background, and KaiC phosphorylation was not known at that time. Therefore, together with our data, we suggest that even their original data (4) may not have demonstrated cyclic KaiC protein synthesis but may have reflected the KaiC phosphorylation rhythm during DD. In addition, the calculated half-life of KaiC in the presence of 400 μg/ml chloramphenicol is ∼10 hours in LL (75), whereas that in DD is >80 hours (Fig. 2, A to C).
