    • Sustained loss of a neoplastic phenotype by brief inactivation of MYC
    • We examined, using the Tet system, if the brief inactivation of an oncogene was capable of inducing sustained tumor regression. We showed that even inactivating the MYC oncogene for as short as a few days in osteogenic sarcomas results in their differentiation. Surprisingly, reactivation of MYC failed to restore their neoplastic properties both in vitro or in vivo, and instead induces apoptosis. These results suggest the possibility that oncogene inactivation can result in a change in the differentiative state of tumor cells and in this new epigenetic context oncogene activation fails to be capable of sustaining tumorigenesis.

    • These results provide compelling in vivo evidence that the MYC oncogene can be restrained from causing tumorigenesis through apoptosis. Blocking the ability of MYC to induce apoptosis was shown to dramatically increase the facility by which MYC activation induced tumorigenesis. In addition, the authors demonstrated that upon MYC inactivation, tumors undergo regression associated with the loss of angiogenesis, changes in expression of cell adhesion molecules and the induction of apoptosis.

    • Impact of p53 loss on reversal and recurrence of conditional Wnt-induced tumorigenesis
    • Hence, oncogene inactivation can induce tumor regression even in the context of the loss of p53 function, but tumors do not exhibit sustained tumor regression. Thus, there are likely to be specific genetic contexts when oncogene inactivation can induce sustained tumor regression. Understanding the mechanism by which loss of p53 function facilitates the relapse of tumors will be very important in the design of therapies that target oncogenes for the treatment of cancer.
