A GFP-PBP2 fusion protein was distributed along the cylindrical sidewalls but not at older cell poles. PBP 2 localized to newly dividing septa but only in the presence of another active Class B enzyme, PBP 3, implying a functional relationship between the two proteins.

Mutations that altered or eliminated these proteins had different effects on cell shape.

By using a fluorescent vancomycin derivative, the authors observed that newly synthesized peptidoglycan formed helical ribbons in the cylindrical side-walls of B. subtilis. Because insertion depended on the Mbl protein (a cytoplasmic actin homolog), the data represent the first hard evidence of a functional relationship between a cytoskeletal protein and peptidoglycan synthesis during cell elongation.

By screening a large collection of strains lacking different combinations of up to seven PBPs, the authors discovered a relationship between the low molecular weight PBPs and cell shape. Shape deformities became apparent only in cells with multiple mutations, and the extent and variation of these aberrations exceeded those of other known mutants, implying that PBPs have a central role in shape creation and maintenance.

Site-directed mutagenesis suggests that this shape-maintenance role depends on DD-carboxypeptidase activity and not on differential localization of PBP 5.
