A GFP-PBP2 fusion protein was distributed along the cylindrical sidewalls but not at older cell poles. PBP 2 localized to newly dividing septa but only in the presence of another active Class B enzyme, PBP 3, implying a functional relationship between the two proteins.

This paper reveals a role for a class B PBP in lateral wall synthesis for two redundant B. subtilis PBPs, homologous to the role of E. coli PBP 2. A double mutant strain was not viable and produced round cells with septation defects.

Immunofluorescence microscopy revealed that the MreB and Mbl proteins form two types of helical filaments along the inner surfaces of the bacterial cytoplasmic membrane. Mutations that altered or eliminated these proteins had different effects on cell shape.

Helical filaments of an Mbl-GFP fusion protein were observed in living cells. The filaments grew with the length of the cell and were split by cell division, and photobleaching demonstrated a continuous exchange of Mbl throughout the filaments.

By using a fluorescent vancomycin derivative, the authors observed that newly synthesized peptidoglycan formed helical ribbons in the cylindrical side-walls of B. subtilis. Because insertion depended on the Mbl protein (a cytoplasmic actin homolog), the data represent the first hard evidence of a functional relationship between a cytoskeletal protein and peptidoglycan synthesis during cell elongation.

The Class A PBPs are the only proteins in B. subtilis known to possess the glycosyl transferase activity required for peptidoglycan strand synthesis. Nonetheless, a strain lacking all four Class A PBPs was viable, produced peptidoglycan with relatively normal structure, and possessed glycosyl transferase activity, indicating the presence of an unknown class of enzymes with this activity.

Two Class A PBPs that are expressed in the developing forespore are capable of producing the first layers of spore peptidoglycan, whereas the majority of the spore peptidoglycan is made by PBPs produced in the mother cell. A double Class A PBP mutant polymerized large disorganized masses of spore peptidoglycan, indicating that the innermost layers serve as a template for synthesis of the outer layers.

By screening a large collection of strains lacking different combinations of up to seven PBPs, the authors discovered a relationship between the low molecular weight PBPs and cell shape. Shape deformities became apparent only in cells with multiple mutations, and the extent and variation of these aberrations exceeded those of other known mutants, implying that PBPs have a central role in shape creation and maintenance.

By combining portions of PBP 5 with homologous segments of PBP 6, the authors identified a stretch of 20 amino acids around the KTG motif of the active site that is required for PBP 5 to maintain a uniform cell shape. Site-directed mutagenesis suggests that this shape-maintenance role depends on DD-carboxypeptidase activity and not on differential localization of PBP 5.

Mol Microbiol 2002, 45:1729-1740. E. coli cells expressing the BolA protein change shape as they enter stationary phase. The authors show that this effect depends on the presence of PBP 5 or PBP 6, suggesting that this normal morphological change is mediated by either of these two PBPs.

de Pedro MA, Young KD, Höltje J-V, Schwarz H: Branching of Escherichia coli cells arises from multiple sites of inert peptidoglycan. J Bacteriol 2003, 185:1147-1152. The authors report the first association of a specific physical feature with sites of branching and morphological defects in bacteria, and suggest that improperly localized patches of inert peptidoglycan create these abnormalities. In addition, the outer membrane over these areas was also stable, implying a close association between peptidoglycan and membrane structure at these sites.
