2 expression of the cdc2 gene. This element apparently binds the p130-E2F-4 complex, as detected by electrophoretic mobility shift assays, and loss of protection of the cdc2 promoter by this complex apparently results in induction of cdc2 gene expression, suggesting a distinct role for a particular E2F complex in transcriptional repression.

These studies, which address the mechanisms by which pRb represses transcription, show that, in transfection experiments, recruitment of pRb to promoters by E2F results in the inhibition of adjacent trans-activators which are able to bind simultaneously to pRb. Thus, tethering pRb to the promoter resulted in repression of c-myc, Elf-1, and PU.1 bound proximally to an E2F site. In vitro binding experiments indicate that pRb may inhibit PU.1 activity by blocking the interaction between PU.1 and TFIID.

In this study, the authors have isolated two new components of the yeast RNA polymerase II holoenzyme, namely SRB10 and SRB11. Interestingly, the two proteins share significant homology with the mammalian Cdk8 and cyclin C proteins (see [40]), and yeast strains deficient for the kinase SRB10-SRB11 exhibit a reduced ability to phosphorylate the carboxy-terminal domain of pol II and a defective transcriptional response to galactose induction in vivo. However, the mutant form of the holoenzyme did not display defects in a reconstituted in vitro transcription assay.
