This interesting report shows that surgical removal of neural tube does not affect the initial expression of myogenic markers, MyoD, myogenin and desmin in somitic cells. Myogenic gene expression, however, is not maintained and is no longer detectable after four days, suggesting that the neural tube may not be required for the initiation of myogenesis, but may be required for the maintenance of the myogenic differentiation program.

Using in vitro co-culture and RT-PCR analysis, the authors demonstrated that myogenesis in pre-segmented mesoderm requires at least two signals derived from the neural tube and floor plate/notochord complex. The data suggest that a signal from ventral midline tissues renders immature somites competent to respond to a subsequent signal from the neural tube. The combination of both signals results in skeletal muscle differentiation in vitro.

The authors propose that Pax-3 plays a role in the specification and/or migration of limb myogenic precursors.
