Glomerular hyperfiltration and the salt paradox in rarly type 1 diabetes mellitus: A tubulo-centric view
-
Vallon V, Blantz RC, Thomson S. Glomerular hyperfiltration and the salt paradox in rarly type 1 diabetes mellitus: a tubulo-centric view. J Am Soc Nephrol 2003; 14:530-537. Why does high salt diet decrease GFR in early diabetes? This review describes the paradoxical relationship between dietary salt and GFR in early diabetes and provides evidence for a primary role for increased proximal tubule reabsorption via a sodium-glucose cotransporter.

Salt-sensitivity of proximal reabsorption alters macula densa salt and explains the paradoxical effect of dietary salt on glomerular filtration rate in diabetes mellitus. Acute hypertension provokes internalization of proximal tubule NHE3 without inhibition of transport activity
-
Yang L, Leong PK, Chen JO, et al. Acute hypertension provokes internalization of proximal tubule NHE3 without inhibition of transport activity. Am J Physiol Renal Physiol 2002; 282:F730-F740. NHE3 redistribution out of the microvilli may not be the central cause of reduced sodium reabsorption if the transporters are first inactivated then targeted for redistribution. This study addressed sodium/hydrogen exchanger transport activity/transporter abundance during hypertension and concludes that NHE3 are not inactivated before transit out of the microvilli.

Responses of proximal tubule sodium transporters to acute injury-induced hypertension
-
Yang LE, Leong PK, Ye S, et al. Responses of proximal tubule sodium transporters to acute injury-induced hypertension. Am J Physiol Renal Physiol 2003; 284:F313-322. A simple renal injection of phenol into one kidney causes immediate hypertension. This paper focuses on NHE3 in the proximal tubule (in both kidneys) and provides the first evidence that NHE3 pool size in the microvilli can be acutely increased by redistribution from a store that may be in the intermicrovillar coated pit region.

-
Yang LE, Zhong H, Leong PK, et al. Chronic renal injury-induced hypertension alters renal NHE3 distribution and abundance. Am J Physiol Renal Physiol 2003; 284:F1056-F1065. The significance of this report is the illustration that while the redistribution of NHE3 to the microvilli in response to phenol injury persists for at least 5 weeks, it is counteracted by a decrease in the total pool size of NHE3, evidence for a compensatory escape mechanisms in the same region of the nephron.

Morris SM, Tallquist MD, Rock CO, Cooper JA. Dual roles for the Dab2 adaptor protein in embryonic development and kidney transport. EMBO J 2002; 21:1555-1564. This paper demonstrates a role for Dab2 in protein absorption in the kidney. A conditional knockout of the Dab2 gene results in proteinuria.

Expression of myosin VI within the early endocytic pathway in adult and developing proximal tubules
-
Biemesderfer D, Mentone SA, Mooseker M, Hasson T. Expression of myosin VI within the early endocytic pathway in adult and developing proximal tubules. Am J Physiol Renal Physiol 2002; 282:F785-F794. How does NHE3 transit from the microvilli to the intermicrovillar cleft? This study illustrated the distribution pattern of myosin VI in the proximal tubule and set the stage for future studies of a role in driving transporters away from the microvilli.

NHE3 serves as a molecular tool for cAMP-mediated regulation of receptor-mediated endocytosis
-
Gekle M, Serrano OK, Drumm K, et al. NHE3 serves as a molecular tool for cAMP-mediated regulation of receptor-mediated endocytosis. Am J Physiol Renal Physiol 2002; 283:F549-F558. This and other cited papers by Gekle and coworkers provide evidence that NHE3 is necessary for efficient receptor-mediated endocytosis of ligands in cultured cells of proximal tubule origin. Here, this group shows, in a model using cellular knockout followed by retransfection, that NHE3 supports receptor-mediated endocytosis and imparts cyclic AMP sensitivity to the process.
