    • Cutting edge: MyD88 is required for resistance to Toxoplasma gondii infection and regulates parasite-induced IL-12 production by dendritic cells
    • This study demonstrates that both DC IL-12 production in response to STAg and host resistance to T. gondii infection depend on the common TLR adaptor molecule MyD88. It thus provides some of the first evidence for a role for TLR signaling in the activation of DCs by a parasite.

    • Leishmania priming of human dendritic cells for CD40 ligand-induced interleukin-12p70 secretion is strain and species dependent
    • The literature on Leishmania infections is complex and often attempts have been made to amalgamate data derived from various in vitro or in vivo studies that have used different species of this parasite. This paper clearly shows that the outcome of the interaction of Leishmania with DCs is very species-dependent and argues therefore that both initial and subsequent responses to these various parasites may be quite dissimilar.

    • DC-SIGN has received much recent attention, not just as a mediator of DC-T cell interactions, but as a pathogen binding receptor. In this paper, the authors describe the first interaction between a protozoan parasite and DC mediated by DC-SIGN. Whether DC-SIGN or structurally related molecules are involved in the pathogenesis of leishmaniasis, however, remains to be determined.

    • In this study the authors identify a new class of molecules that trigger DC maturation and show that TLR-2 is not sufficient for cellular binding. The latter observation is important in that it provides a framework for cooperation between many cellular receptors involved in pathogen recognition and subsequent TLR-mediated signaling.
