Multivesicular release at climbing fiber-Purkinje cell synapses

Following up on their earlier work [43], the authors demonstrate that the peak concentration of glutamate at climbing fiber synapses on Purkinje cells is dependent upon the release probability of synaptic vesicles, and propose that the simultaneous release of multiple vesicles is the underlying mechanism.

A single packet of transmitter does not saturate postsynaptic glutamate receptors

This study shows that the AMPAR- and NMDAR-mediated synaptic events are potentiated by presynaptic glutamate infusion in the giant nerve terminal, the calyx of Held, which suggests that the level of cytoplasmic transmitter can affect the loading of transmitter into synaptic vesicles.

A developmental switch in neurotransmitter flux enhances synaptic efficacy by affecting AMPA receptor activation
-
The authors of this study and Choi et al. [41] report that the slow flux of glutamate is unable to activate AMPA receptors, while able to activate NMDA receptors in the same synapse, which indicates that the temporal profiles of transmitter flux are important in determining postsynaptic receptor activation.

The expression of vesicular glutamate transporters defines two classes of excitatory synapse
-
This study reports the identification of a second type of vesicular glutamate transporter, VGLUT2 and demonstrates the complementary expression of VGLUT1 and VGLUT2. The distinct distribution patterns of these transporters suggest that they might define two classes of excitatory synapse.

Altered presynaptic vesicle release and cycling during mGluR-dependent LTD
-
Using fluorescent probe FMI-43 uptake and release during exocytosis and endocytosis as a tool, the authors demonstrated that when the strength of synaptic transmission is reduced during LTD, presynaptic terminals are capable of taking up FM dye, but incapable of releasing FM dye. They proposed that the reduction of synaptic transmission might be due to the alteration of kinetics of presynaptic transmitter release.
