A genetic locus determining susceptibility/resistance to West Nile virus infection in mice was mapped to chromosome 5, where a cluster of genes encoding a family of IFN-inducible proteins (2′-5′-oligoadenylate synthetases) was found to be the most prominent candidate. Nucleic acid sequence comparison of the three isoforms of the gene showed a mutation causing a stop codon and thus a truncated protein was associated with susceptibility. The implication is that, in animal models at least, a point mutation in a single gene may determine host susceptibility to West Nile virus infection.
