    • Efficient specific DNA binding by p53 requires both its central and C-terminal domains as revealed by studies with high-mobility group 1 protein
    • In this study, the authors analyse the ability of p53 to bind to bended and supercoiled DNA and provide a demonstration that, under these conditions, the carboxyl terminus of p53 is important for promoting DNA binding. Both the sequence-specific DNA-binding domain and the carboxyl terminus therefore cooperates to recognise sequence and structure in genomic DNA.

    • Chromatin immunoprecipitation analysis fails to support the latency model for regulation of p53 DNA binding activity in vivo
    • Using the chromatin immunoprecipitation (ChiP) approach, the authors provide clear evidence that, in vivo, p53 is bound to its target promoters with the same affinity in the absence or in the presence of DNA damage. This suggests that activation of p53-mediated transcription does not require increased DNA binding.

    • SUMO-1 protease-1 regulates gene transcription through PML
    • The authors identified SuPr-1, a SUMO-1-specific protease, as an activator for c-Jun-dependent transcription that is independent of c-Jun phosphorylation. They provide evidence that SuPr-1 acts on transcriptional activation in part in a promyelocytic leukaemia protein (PML)-dependent manner by altering the subcellular distribution of the transcriptional regulators PML, CBP and Daxx. This paper indicates an important transcriptional regulatory function of SUMO proteases.

    • This paper is one of the few studies dedicated to analyse the behaviour of different promyelocytic leukaemia protein (PML) isoforms. The authors demonstrate that, in human normal fibroblasts, only PML IV is able to induce premature senescence and to activate p53. This effect, while mediated by interaction between PML, CBP and p53, seems not to require the integrity of nuclear bodies.
