The results described allow the identification of a number of steps at which lipid-mediated transfection is inefficient. According to their data, a large amount of DNA is delivered to the cell, a small percentage of that is released from the endosome, and a small percentage of that makes its way from the cytoplasm to the nucleus where it is transcribed. The inefficiency of each step suggests that specific attention must be paid to developing ways to overcome each of the different barriers.

The authors investigated which biomolecules interfere and mediate release of DNA from a complex with cationic liposomes. They demonstrated that anionic lipids normally found on the cytoplasmic-facing monolayer of cell membranes can very potently displace DNA from the cationic liposome/DNA complex. On the basis of their results, the authors proposed a mechanistic model to explain the release of DNA from the cationic liposome complex in cells.

This work provides important insights into the interactions established between DNA and cationic liposomes as well as into the structure of the resulting lipid/DNA complexes. By carrying out a combined in situ optical microscopy and X-ray diffraction study, the authors conclude that the complexes consist of a higher ordered multilamellar structure with DNA sandwiched between cationic lipid bilayers. These findings may lead to a better understanding of the important structural parameters relevant to transfection efficiencies in gene therapy.

It was shown that an amphiphile (#67) consisting of a cholesterol anchor linked to a spermine headgroup in a 'T-shape' configuration is as efficient for gene transfer to the lung as adenovirus vectors without eliciting a host immune response.

Incorporation of cholesterol in multilamellar liposomes significantly increased the expression levels of different trangenes, which was attributed to a prolonged circulation time of injected lipoplexes and increased uptake and retention in tissues.

Biochim Biophys Acta (1999) in press. The relationship between the characteristics of the lipoplexes, their mode of interaction with cells and their ability to mediate transfection was investigated. By monitoring the dequenching of a non-exchangeable fluorescent probe embedded in the liposome membrane, the authors assessed the effects of the lipid/DNA charge ratio, endocytosis inhibitors and serum on membrane lipid mixing. The authors conclude that no correlation between fusion with the plasma membrane and transfection can be established. This lack of correlation was also reported by others [61].

This report describes a new methodology that leads to the formation of stable cationic lipid/DNA complexes that maintain the same transfection efficiency for up to three months and that can be concentrated with no loss in activity. Briefly, this new procedure involves the solubilization of cationic liposomes with 1% octytglucoside and complexation of plasmid DNA with the lipid in the presence of detergent. The stable complexes were obtained after removal of the detergent by dialysis. This study, as well as that presented in reference [54•], provides an important contribution to the development of alternative approaches to the use of cationic liposomes as gene carriers.

This paper describes a high efficiency, non-viral gene transfer method employing cationic liposomes plus a receptor ligand. This protocol results in 100% transfection of cells when lipofectin is associated with transferrin. The enhancement of liposome-mediated gene transfer efficiency and expression by transferrin was shown to vary among different cationic liposomes. Similar strategies were also reported in references [28,66•], although such a high efficiency of transfection could not be reproduced.

A combination strategy involving cationic liposomes to condense and carry DNA, transferrin to facilitate cellular uptake, and the pH-sensitive peptide GALA to promote endosome destabilization, resulted in significant expression of a luciferase gene in primary human macrophages. Transfection of these cells was dependent on the degree of differentiation of the cells. The quaternary complexes of cationic liposomes, DNA, transferrin and GALA exhibited a net negative charge, which may obviate a limitation of cationic synthetic vectors in vivo. The lack of cytotoxicity and the expected lack of immunogenicity of these complexes may render them useful for gene delivery to macrophages in vivo. In this context see also references [28,65••].

Although the authors demonstrate that the use of such strategy leads to a significant enhancement of transgene expression, it remains to be clarified whether this effect resulted from an increase of DNA delivery into the nucleus.

This work describes the potential of using cationic liposomes composed of a cationic lipid (DOTAP, DOGS, DDAB), a neutral lipid (DOPE) and a phospholipid derivative of poly(ethylene glycol) (PEG-PE) to prepare complexes with oligonucleotides (ODN) aimed at enhancing their biological activity. It was demonstrated that modification with PEG-PE not only increases particle stability of cationic liposome-ODN complexes but also allows extraordinarily high loading of ODN, resulting in neutral or negatively charged particles. Of great relevance is the finding that conjugation of antibody fragments recognizing the HER2 receptor to such cationic liposome-ODN complexes triggers efficient delivery of ODN to their sites of action, resulting in enhanced biological activity of antisense ODN in a target-specific manner.
