Plant J 2002, 29:225-235.This study establishes the physiological importance of AtTPS1. This gene is required for embryo maturation and the induction of marker genes for storage-reserve deposition. This work provides evidence that growth arrest in the embryo of tps1 mutants is caused in part by the increased supply of sucrose that occurs as the seed matures. Disruption of TPS1 in S. cerevisiae also prevents growth on sugars [6••] but the mechanism established in yeast does not appear to fit Arabidopsis

The E. coli OtsA protein has a twin-Rossmann fold with glucose-6-phosphate binding to the amino-terminal domain and uridine-5-diphosphoglucose to the carboxy-terminal domain, catalysis occurs at the interface between the domains

    • Paul M, Pellny T, Goddijn O: Enhancing photosynthesis with sugar signals. Trends Plant Sci 2001, 6:197-200. A recent review in which the authors report that elevating T-6-P levels by overexpressing E. coli TPS increases photosynthetic rate (per unit leaf area), whereas reducing T-6-P levels by overexpressing E. coli TPP reduces photosynthetic rate. These findings indicate a correlation between T-6-P levels (rather than trehalose levels) and changes in plant physiology

Proc Natl Acad Sci USA 2002, 99:15898-15903. These authors report the overexpression of the E. coli trehalose biosynthetic genes otsA and otsB as a single gene fusion. The transgenic plants accumulated trehalose but at levels well below that required for trehalose to act as an osmoprotectant under abiotic stress conditions. Increased trehalose content correlates with an overall increase in soluble carbohydrates, consistent with a proposed role for trehalose metabolism in the regulation of carbon metabolism

Biochem J 2002, 366:63-71. The authors show that AtTPS1 contains an amino-terminal extension, which may act as an inhibitory domain that allows the modulation of TPS activity.
