Plant Physiol 2001, 125:1548-1553. The authors transformed tobacco BY2 cells and selected a cell line that expressed high levels of TetR protein. They evaluated the capacity of this cell line to suppress the GFP target gene in the absence of tetracycline and demonstrated that it is an excellent recipient cell line for recovery of clonal cell lines in which the regulation of the introduced gene is highly controlled.

The authors optimized the system by cloning the activator and target genes in different orientations. This system is suitable for both laboratory and field use.

The inducible expression of the Ms45 protein in the tapetum cell layers restored the fertility of male sterile ms45 maize. This is an excellent application of a chemically inducible system for commercial use.
