Transplantation and engraftment of wild-type bone marrow into tumor-resistant Id-mutant mice results in restoration of tumor angiogenesis and growth. These studies demonstrate that comobilization of VEGFR1 + CEPs and VEGFR2+ hematopoietic cells are essential for tumor angiogenesis. The primary angiogenic defect in Id-mutant mice is the impaired capacity of CEPs and hematopoietic cells to mobilize and incorporate into tumor vasculature.

Cell 2002, 109:625-637. •• Shows the mechanism by which bone marrow-derived stem and progenitor cells are recruited from the bone marrow microenvironment and mobilize to peripheral circulation. Physiologic stressors, including bone marrow suppression or elevation of angiogenic factors, induce MMP-9, resulting in the release of soluble Kit ligand, driving proliferation and mobilization of stem and progenitor cells.

Nat Med 2002, 8:841-849. • Defines the mechanism by which angiogenic factors, including PIGF and VEGF, recruit hematopoietic cells from the bone marrow to the circulation. VEGFR1 is expressed on hematopoietic stem and progenitor cells and conveys signals that promote motility and MMP-9-mediated release of soluble Kit ligand, thereby directing mobilization of these cells to the peripheral circulation.
