    • For misinsertion experiments, relatively high dNTP concentrations have to be applied to detect nucleotide incorporation (Ref. [6b]). Higher dNTP concentrations cause nucleobase-dependent extension of DNA products as a result of misinsertion caused by mismatched extension. This provokes the generation of longer reaction products than expected through single nucleotide insertion alone (see Figure 2, lanes 1-4).
