Randomized comparison of megestrol acetate versus dexamethasone versus fluoxymesterone for the treatment of cancer anorexia/cachexia
-
Loprinzi CL, Kugler JW, Sloan JA, et al. Randomized comparison of megestrol acetate versus dexamethasone versus fluoxymesterone for the treatment of cancer anorexia/cachexia. J Clin Oncol 1999; 17:3299-3306. This large randomized trial demonstrated that megestrol acetate and dexamethasone are equivalent in their orexigenic effects at the doses prescribed and that the androgen fluoxymesterone has relatively little palliative effect. This study provides the rationale, based on the toxicity profile of each agent, for treating patients with a limited life expectancy with dexamethasone and with a slightly longer life expectancy with megestrol acetate.

Randomized clinical trial of adenosine 5′-triphosphate in patients with advanced non-small cell lung cancer
-
Agteresch HJ, Dagnalie PC, der Gaast A, et al. Randomized clinical trial of adenosine 5′-triphosphate in patients with advanced non-small cell lung cancer. J Natl Cancer Inst 2000; 92:321-328. This clinical trial tested a novel agent, ATP, in the treatment of cancer-associated anorexia and weight loss in patients with advanced non-small cell lung cancer. The favorable results in this study suggest that ATP deserves further testing.

Intratumoral injection of oligonucleotides to the NFkappaB binding site inhibits cachexia in a mouse tumor model
-
Kawamura I, Morishita R, Tomita N, et al. Intratumoral injection of oligonucleotides to the NFkappaB binding site inhibits cachexia in a mouse tumor model. Gene Ther 1999; 6:91-97. By testing a decoy of oligonucleotides that block NF-κB transactivation of gene expression of cytokines, this study introduces a novel approach to the palliation of cancer-associated anorexia and weight loss.
