An almost complete analysis - for the first time - of a eukaryotic protein complex by extensive two-hybrid screening. In addition to the identification of protein contacts between the individual RNA polymerase III subunits, the authors also find a novel subunit. In combination with crystallographic studies these results should permit a deeper insight into the structure and, therefore, mechanism of transcription.

Besides screening a number of known splicing proteins, the authors also use preys they identified as second-round baits and thereby were able to generate a fairly complicated network of protein-protein interactions.

This is the most extensive, systematic study of protein-protein interactions published so far. The study involves more than a thousand proteins and generated an equal number of interactions. A comparison of library and array screen shows that the former is much faster but yields fewer interactions.

A biochemical genomics approach for identifying genes by the activity of their products

Before the study of Martzen et al. appeared, biochemical experiments were thought to be too complicated to be done on a genomic scale. These authors for the first time purified all 6000 yeast proteins as GST fusions and tested them for biochemical activities. This is potentially an excellent way to verify two-hybrid interactions and may even allow to quantify protein-protein interactions.
