2+ Atpase gene cause deafness in deafwaddler mice
-
The deaf waddler mouse mutant is deaf and displays vestibular/motor imbalance. Mutations have been detected in the gene encoding PMCA2: in one of the mutants, an A→G transition causes a Gly→Ser substitution at the highly conserved position in the pump sequence. In another mutant, a 2 bp deletion causes a frameshift that produces a truncated pump. PMCA2 is localised to stereocilia and the basolateral wall of hair cells of the cochlea in wild-type mice, but is not detected in mice with the mutation producing the truncated pump.

PMCA2-null mice grow more slowly than heterozygous and wild-type mice, have difficulties in maintaining balance, and are deaf. Histological analysis of the cerebellum and inner ear shows an increased number of Purkinje neurons, a decreased thickness of the molecular layer, and a range of abnormalities in the inner ear (vestibular system and the organ of Corti).
