    • 2+ Atpase gene cause deafness in deafwaddler mice
    • The deaf waddler mouse mutant is deaf and displays vestibular/motor imbalance. Mutations have been detected in the gene encoding PMCA2: in one of the mutants, an A→G transition causes a Gly→Ser substitution at the highly conserved position in the pump sequence. In another mutant, a 2 bp deletion causes a frameshift that produces a truncated pump. PMCA2 is localised to stereocilia and the basolateral wall of hair cells of the cochlea in wild-type mice, but is not detected in mice with the mutation producing the truncated pump.

    • 2+ might induce a rearrangement of the TMSs, and is subsequently transmitted to the cytoplasmic domain through a structural change in the stalk. (However, see the text for the alternative possibility that the structural change reflects the binding of vanadate instead.) Significant changes around the putative ATP-binding site might be explained by the segmental movement of the cytoplasmic domains.
