Acetylcholine-induced nitrite production and mRNA for eNOS were significantly increased in the vasculature from exercised dogs compared with control animals. Elevated eNOS expression leading to increased production of NO may explain the improvement in endothelial function observed.

After chronic exercise, vasodilation was increased in response to bradykinin (NO agonist) and was inhibited after addition of L-N-monomethyl-L-arginine (NO antagonist). No difference was observed in dilation in response to adenosine or nitroprusside. These data suggest that endothelium-dependent vasodilation mediated by the production of NO is improved after exercise training.

Using venous occlusion plethysmography, forearm blood flow was measured in response to hand-grip exercise before and after intra-arterial infusion of the NO inhibitor L-NMMA in patients with congestive heart failure. Flow was significantly diminished in normal subjects compared with patients with congestive heart failure, indicating that NO-mediated vasodilation is impaired in patients with congestive heart failure.
