This is the first paper detailing the effects on a whole organism of the knock-out of the linker histone H1 gene that encodes a protein that contains a metazoan-like tripartite structure. In Ascobolus, the authors show that H1 is not required for establishing methylation-dependent repression of selected genes, and that its loss results in hypermethylation of previously methylated chromosomal regions. Strains harboring silenced H1 exhibit a shortened life span. On the level of chromatin, nucleosome repeat is not changed in cells lacking H1, but, as expected, the chromatin is more accessible to micrococcal nuclease. More importantly, H1 binds in a similar way in vivo to methylated versus unmethylated DNA, resolving a recently contentious point from in vitro experiments

The authors report the cloning and identification of qde-1, a gene encoding an RNA-dependent RNA polymerase. This is the first isolated cellular component known to be involved in quelling, a post-transcriptional gene silencing (PTGS) mechanism operating in Neurospora. The presence of qde-1 homologues in a variety of species of plants, animals and fungi supports the idea that a conserved PTGS mechanism may operate in eukaryotes
