Scavenger receptor BI (SR-BI) mediates free cholesterol flux independently of HDL tethering to the cell surface. J Lipid Res 1999; 40:575-580. By comparing the closely related receptors SR-BI and CD36, the authors show that SR-BI is more efficient than CD36 in promoting free cholesterol efflux. In these tissues holoparticle endocytosis of HDL seems to be the main pathway for cholesteryl ester uptake.

    • 12 receptor, cubilin, is a high-affinity apolipoprotein A-I receptor facilitating endocytosis of high-density lipoprotein. Nat Med 1999; 5:656-661. In this paper cubitin is shown to be a high affinity receptor for apolipoprotein AI and to mediate the endocytosis of HDL in yolk sac cells. In-vivo data indicate an important role of renal cubilin in the metabolism of apolipoprotein AI.

Studies in apoA-II and hepatic lipase double knockout mice. J Lipid Res 1999; 40:1064-1070. Using apolipoprotein AII and hepatic lipase-deficient mice, the authors demonstrate that apolipoprotein AII is a physiological inhibitor of hepatic lipase and that this is at least a part of the mechanism whereby apolipoprotein AII maintains HDL cholesterol levels.

This paper reports a third lipase expressed in endothelium. Overexpression of the lipase in mice decreases the level of HDL. The novel lipase is suggested to have an important function in lipoprotein metabolism and vascular biology.

J Clin Invest 1999; 104:R25-R31. In this paper the authors provide functional data demonstrating that the ABC1 protein regulates cellular cholesterol efflux, which may be important for the size of HDL and thereby HDL clearance. Consistent with similar reports this year from other laboratories, mutations in the ABC1 transporter are shown to be responsible for Tangier disease.
