Mechanisms involved in the regulation of free fatty acid release from isolated human fat cells by acylation stimulating protein and insulin. J Biol Chem 1999; 274:18243-18251. An important extension of the potential physiologic significance of the ASP pathway is provided. This article establishes that ASP is an important determinant of fatty acid release from adipocytes, as well as a critical determinant of their deposition. Furthermore, the actions of ASP are complementary to those of insulin in regulating fatty acid release. New insights are provided into the signal transduction mechanisms that are responsible for these effects of ASP.

J Biol Chem 1999; 274:19429-19433. In this study, triglyceride clearance was not delayed in ASP knockout mice, or was plasma apoB elevated. Accordingly, the authors concluded that the ASP pathway is not physiologically relevant.

In our study, plasma triglyceride clearance was delayed in the ASP knockout mice, an effect that was particularly evident in the males. Intraperitoneal injection of human ASP significantly accelerated triglyceride clearance in the knockout male animals.

Am J Physiol Endocrinol Metab 1999; 277:E474-E480. The data from this study demonstrate that intraperitoneal injection of human ASP significantly accelerated triglyceride clearance, particularly in those animals in which it was not already accelerated at baseline. It represents clear evidence that the effect of ASP on triglyceride clearance, as distinguished from that on fatty acid trapping, is evident only when the clearance is already limited.
