By in vitro binding experiments, the authors show that components of the TOM complex differ in their affinities for mitochondrial-targeting sequences. On the basis of this observation, the authors propose the 'acid chain' hypothesis, according to which preproteins interact in a sequential order with binding sites of increasing affinity which leads to a translocation of the preproteins through the TOM channel.
