    • TRF2 protects human telomeres from end-to-end fusions
    • ••] demonstrate the important role of two myb-related telomere binding proteins in these processes. TRF1 negatively regulates telomere length; inhibition of TRF1 causes telomeres to lengthen. TRF2 serves an important structural role at the telomere. Dominant-negative TRF2 causes rapid chromosomal fusion, indicating that TRF2 normally suppresses fusion of chromosomes at their telomeres.

Moss T. de Lange Mammalian telomeres end in a large duplex loop Cell 97 1999 503 514 One of the main functions of telomeres is to prevent recognition of the chromosome ends as a double-strand break. This report shows that mammalian telomere ends are not exposed as linear structures but instead loop back to interact with the more proximal portion of the telomere. Purified TRF2 alone can facilitate this looping event, suggesting a model in which loss of TRF2 function might cause exposure of this end, making it recognizable as a double-strand break.

Interestingly, they differ in their conclusions on the effect of TERT telomere length. Telomeres shortened despite TERT expression in the latter report, suggesting a possible role for telomerase in protectively capping the telomere end.
