A Salmonella virulence protein that inhibits cellular trafficking

This paper elegantly demonstrates that the bacterial protein SpiC secreted by a type III secretion system into the host cell cytoplasm specifically inhibits fusion of Salmonella-containing phagosomes with lysosomes and endosomes, and interferes with trafficking of vesicles devoid of the microorganism. Furthermore, it was shown that the ability to interfere with intracellular trafficking is essential for Salmonella pathogenesis.

This paper describes the differential expression of listerolysin O (LLO) and ActA by intracellular (cytoplasmic) versus extracellular (broth-grown) L. monocytogenes. Reporter gene fusions of lacZ and cat are used to document the expression levels of the hly and the actA gene during infection. It is shown that the levels of induction of the two genes are strikingly different despite the dependence of the expression of both genes on the transcriptional activator PrfA.

Using RT-PCR and green fluorescent protein reporter gene fusions to different L. monocytogenes promotors, differential gene expression in different intracellular compartments (phagosome and cytosol) is documented. Additionally, differences between intracellular expression of listerial genes in macrophages, epithelial cells, endothelial cells, and hepatocytes are also demonstrated pointing to complex regulatory networks controlling intracellular gene expression.

Pathogen-induced apoptosis is a widely used bacterial strategy during infection. In contrast to other bacterial pathogens, Rickettsia rickettsii actively inhibits host cell apoptosis in a NF-κB-dependent pathway allowing the bacteria to remain in the infected endothelial cell.
