    • A single myosin head moves along an actin filament with regular steps of 5.3 nanometres
    • This paper describes a new assay with which individual S1 molecules can be captured and directly manipulated using a scanning probe. Using this assay, it is shown that an S1 molecule continuously moves along an actin filament with regular steps of approximately 5.3 nm and undergoes five steps to produce a maximum displacement of 30 nm during a single biochemical cycle of ATP hydrolysis.

    • The authors measured the rising phases of displacements for unconventional myosin 1, which was much slower than skeletal myosin and thus can be followed by optical trapping nanometry. They observed two substeps with a size of 5-6 nm in the rising phase.

    • Simultaneous observation of individual ATPase and mechanical events by a single myosin molecule during interaction with actin
    • The authors combined a technique for single molecule imaging with optical trapping nanometry to simultaneously observe the individual mechanical and ATP turnover events of a single myosin head. They showed that each overall displacement corresponded to a single ATP turnover event to that of chemical ones, suggesting that the chemical energy of ATP hydrolysis is stored in the head and gradually used later.
