Novel non-heme iron center of nitrile hydratase with a claw setting of oxygen atoms
-
The crystal structure of nitrile hydratase at 1.7 Å resolution, combined with mass spectrometry data, revealed the structure of the non-heme iron catalytic center in the nitrosylated state. Two cysteine residues coordinated to the iron were found to be post-translationally modified to cysteine-sulfenic (Cys-SOH) and cysteine-sulfinic acids (Cys-SOOH). Together with another oxygen atom of the serine ligand, these groups form a 'claw' setting of oxygen atoms capturing an NO molecule

Kato Y., Ooi R., Asano Y. A new enzymatic method of nitrile synthesis by Rhodococcus sp. strain YH3-3. J Mol Catal B. 6:1999;249-256. The authors describe a new enzymatic nitrile synthesis using the aldoxime dehydratase from Rhodococcus sp. YH3-3. 3-cyanopyridine production was achieved with 72.5%, 19.7% and 2.36% yields from the E-pyridine-3-aldoxime analogs O-acetyl-E-pyridine-3-aldoxime, Z-pyridine-3-aldoxime and E/Z-pyridine-3-aldehyde hydrazone, respectively.
