Mutation detection of PKD1 identifies a novel mutation common to three families with aneurysms and/or very-early-onset disease

22 Watnick T, Phakdeekitcharoen B, Johnson A, et al. Mutation detection of PKD1 identifies a novel mutation common to three families with aneurysms and/or very-early-onset disease. Am J Hum Genet 1999; 65:1561-1571. An important study for two reasons. First, due to the improved methodology for mutation detection in the reduplicated portion of PKD1, and second because common germline mutation with severe phenotype suggests the possibility of genotype/phenotype correlation, perhaps based on effects of partial protein product.

Major findings include the following: that hypertension is a markedly less frequent complication in PKD2 than PKD1, PKD2 is significantly less severe in women but that this is not the case for PKD1, the late age in diagnosis in PKD2 confirms the possibility of under-ascertainment, there may be a decrease in survival in PKD2 that is independent of renal complications.

Aberrant splicing in the PKD2 gene as a cause of polycysitic kidney disease

29 Reynolds DM, Hayashi T, Cai Y, et al. Aberrant splicing in the PKD2 gene as a cause of polycysitic kidney disease. J Am Soc Nephrol 1999; 10:2342-2351. Two major points from this paper are that genomic mutation detection without confirmation at the level of the gene transcript can be deceptive, and lymphoblasts from PKD2-affected individuals express half normal levels of polycystin-2 (potential of diagnostic use).

The rat Pkd2 protein assumes distinct subcellular distributions in different organs
-
38 Obermuller N, Gallagher AR, Cai Y, et al. The rat Pkd2 protein assumes distinct subcellular distributions in different organs. Am J Physiol 1999; 277:F914-F925. This first immunolocalization of polycystin-2 in adult rat tissues suggests differential subcellular distributions in different tissues. It raises the possibility that polycystin-2 has different functions in different cell types and that binding partners will likely also be tissue specific.

40 Cai Y, Maeda Y, Cedzich A, et al. Identification and characterization of polycystin-2, the PKD2 gene product. J Biol Chem 1999; 274:28557-28565. The authors established that polycystin-2 is expressed in the endoplasmic reticulum of cells in culture. Furthermore, the study identifies a 34 amino acid stretch in cytoplasmic tail that is essential to this localization. In light of the putative interaction of polycystin-1 and -2, this work raises the possibility that polycystin-1 or another binding partner is required to relocalize polycystin-2 to the plasma membrane or that PKD1 and PKD2 interact across separate membranes.

A polycystic kidney disease gene homolog required for male mating behavior in Caenorhabiditis elegans
-
46 Barr MM, Sternberg PW. A polycystic kidney disease gene homolog required for male mating behavior in Caenorhabiditis elegans. Nature 1999; 401:386-389. The C. elegans homologue of polycystin-1 is a mechano-or chemosensory signal transducer that co-localizes with PKD-2. Powerful C. elegans genetics should allow ready identification of additional players in this signaling pathway and provides ready in-vivo assay.
