The authors describe a novel family of secreted Wnt antagonists that block Wnt signaling upstream of Dishevelled. Dickkopf-1 (dkk-1) may operate outside of the cell similarly to FRPs, however it is not known whether it binds Wnts directly or has different molecular targets. Together with FRPs, chordin, noggin and cerberus, dkks appear to control cell fates by attenuating rather than activating signal transduction.

The targets of PP2A in the Wnt pathway remain to be elucidated.

Sakanaka C, Williams LT: Functional domains of axin. J Biol Chem 1999, 274:14090-14093. This report shows that the carboxy-terminal domain of Axin is required for protein oligomerization and activity. Axin constructs lacking this domain failed to suppress TCF-dependent transcription in SW480 cells with an APC mutation, yet a similar construct may be active in Xenopus embryos (see [41•]).

Science 1998, 280:1274-1277. This paper shows that two related Wnts use different molecular pathways. Wnt3a and Wnt7a are coexpressed in dorsal ectoderm of the chick limb bud. Both proteins belong to the same class of highly transforming Wnts but induce different target genes in the limb. The effect of Wnt3a, but not that of Wnt7a, was mimicked by and required active TCF/β-catenin signaling.
