The authors identify truncated, soluble forms of Delta in extracts of Drosophila tissues and cultured cells (see also [15, 16]). They show that the accumulation of processed Delta requires Kuzbanian, a matrix metalloprotease that is required for normal Notch signaling, and that processed Delta can bind to Notch. They also present some evidence that processed Delta can activate Notch but the assay employed is ill-defined and more experiments are needed to confirm this.

The authors report that mutation of nubbin in the Drosophila wing causes ectopic expression of Notch target genes away from the D-V boundary and that over-expression of nubbin can repress the expression of Notch target genes. They go on to show that at least in the case of vestigial, this effect appears to be generated by binding of Nubbin protein to an enhancer that is a direct target of Notch signaling.

They also show that expression of a Notch target gene, Hes-5, is disrupted in L-Fng mutants, further strengthening the argument that L-Fng is affecting Notch signaling in mammals. They report that expression of Notch ligands is more diffuse in L-Fng mutants, and present three possible models to explain the L-Fng mutant phenotype.

The authors describe the expression of fringe, Serrate, and Delta in the eye imaginal disc. They also describe the phenotypes generated by fringe mutants, and those generated by ectopic expression of Fringe, Serrate, Delta, and activated-Notch. They show that the effects of Serrate and Delta are modified by Fringe just as they are in the wing. They show that Notch is activated along borders of Fringe expression in the eye, and that Notch activation defines a D-V midline cell fate. In addition to promoting the expression of D-V midline genes, they show that Notch activation also promotes eye growth and influences ommatidial chirality.

They also show that Notch target genes are expressed along the D-V midline, and that fringe mutant clones can influence ommatidial chirality.

By analyzing clones of cells mutant for fringe, Serrate and Delta, or Notch, and clones of cells ectopically expressing Fringe, Serrate, Delta, or activated-Notch, they show that local activation of Notch is necessary and sufficient to promote the formation of joints between leg segments. They report that fringe, Serrate, and Delta are each expressed in segmentally repeated rings in both leg and antennal imaginal discs, and that the establishment of this segmentally repeated expression is regulated downstream of Wingless and Dpp. They also show that Notch activation is both necessary and sufficient to promote leg growth, and they identify target genes regulated downstream of Notch that are required for normal leg development. On the basis of these results, they propose a regulatory hierarchy for the segmentation and growth of the leg.

They report that Serrate and Delta are expressed in segmentally repeated rings in leg imaginal discs but disagree with [36•] as to the expression and requirements for fringe. They also show that Notch activation is necessary for normal leg growth, and show that reporter gene constructs that respond to Notch signaling, E(spl)mβ-CD2 and bib-lacZ, are expressed in distal cells of each leg segment. They also describe the temporal profile of E(spl)mβ-CD2 and bib-lacZ expression, and their results imply that leg segmentation occurs progressively over the course of the third larval instar.
