In this note, the following equation was used in order to calculate the vertical (z axis) resolution (fwhm): fwhm [/mi] = c{[2n/(M(N.A.))]-Pfdig] + nA(EXC)/((N.A.) × (N.A.)) × exp{-[2(N.A.)/(A(EXC)-Af)]P[dig]}. Here c is 1.41 × A(EM)/A(EXC) for the fluorescent mode and 1 for the reflection mode, A(EM) is the wavelength of emitted light, A(EXC) is the wavelength of the incident light, N.A. is the numerical aperture of the objective, M is the magnification of the objective, n is the refractive index of medium, andP[dig] is a special numerical value assigned to the pinhole used.
