This study definitively establishes a role for Hwp1 in adhesion of C. albicans germ tubes to buccal epithelial cells (BECs) by the use of hwp1 null mutants and complemented strains. The amino terminus of Hwp1, expressed as a recombinant protein, and germ tube surfaces of C. albicans displayed TGase substrate activity. Adhesion of germ tubes and recombinant Hwp1 to BECs was resistant to thermal denaturation in the presence of SDS and was susceptible to inhibitors of transglutaminase. The study provides new insight into how C. albicans colonizes host surfaces.

[30••], this study also demonstrates a relationship between O-glycoslyation and adherence, but extends the relationship by showing that CaMNT1p adds a second mannose residue to singly mannosylated hydroxy amino acids. The results show that O-linked chains longer than a single mannose are important for adhesive properties of cells.
