After cryofixation and Epon embedding the three major nucleolar components were also identified in yeast. By in situ hybridization and immunogold-electron microscopy evidence is provided that the functional organization of the yeast nucleolus is comparable to that of metazoan cells (see also [8•]).

Nucleoli and coiled bodies share several molecular components such as Nopp140, fibrillarin and U3 snoRNP. Transient expression of the highly evolutionarily conserved carboxyl terminus of Nopp140 in mammalian cells caused redistribution of endogenous Nopp140 and fibrillarin to the nucleoplasm and disintegration of the coiled bodies. Interestingly, the structural integrity of the Nopp140 and fibrillarin-deficient nucleoli was not affected as shown by phase-contrast microscopy. The authors propose that the dominant-negative effect of the partial Nopp140 construct is due to its binding to an as yet unknown nucleoplasmic factor. Titration of this factor could impair the function of endogenous Nopp140 in shuttling other proteins and snoRNPs to the nucleolus and coiled bodies.

The authors describe the localization of several maize snoRNAs which are all generated from a common precursor. By in situ hybridization they show that the different snoRNAs localize to different nucleolar regions reflecting their involvement in early and late pre-rRNA processing steps. Most interestingly, probes for the intergenic spacer regions led to the detection of snoRNA precursors both in nucleoli and coiled bodies. The data are compatible with the view that polycistronic precursors are targeted via the coiled bodies to the nucleoli where they are processed and assembled into mature snoRNPs.

Sequence requirements for nucleolar localization of the three box C/D snoRNAs U3, U8 and U14 were identified by injection of fluorescein-labeled mutated snoRNAs into Xenopus oocyte nuclei and immunofluorescence [36•-38•]. Control experiments with oocytes in which endogenous snoRNAs were depleted by prior injection of antisense oligonucleotides demonstrated that the injected wild-type snoRNAs localized correctly to the nucleoli and were able to rescue pre-rRNA processing. Boxes C and D were identified as primary nucleolar localization elements.
