Science 1998, 281:838-842. Deletion of the gene for DARPP-32 in mice causes a dramatic loss in the response of neostriatal neurons to dopamine. The authors demonstrate that the DARPP-32/PP1 cascade is an obligatory component, along with PKA, in dopamine signaling. This is one of two published studies (see also [19•]) demonstrating a specific physiological function that can be attributed to PP1 in intact animals.

Previous studies using inhibitors of PSPs implicated PP2A in the regulation of a number of protein kinases. The data reported here support those findings and describe a mechanisms through which PP2A could act as a ubiquitous regulator of kinases. Similar complexes of PP2A and neuronal protein kinases are likely to exist.

This paper shows that induction of tau hyperphosphorylation with okadaic acid correlates with the loss of stable microtubules and degeneration of axons in cultured neuronal cells. The results demonstrate that PSPs play a constitutive role in dephosphorylation of tau. Suppression of phosphatase activity leads to phosphorylation and functional changes similar to those in pathological states, such as in Alzheimer's disease.
