This paper describes the first cloning and characterisation of catalytic subunits of telomerase. The gene for a Euplotes aediculatus protein (p123), previously shown biochemically to copurify with telomerase activity, was cloned and sequenced and found to contain reverse transcriptase motifs. The Saccharomyces cerevisiae homologue of this gene was found to encode Est2p, a protein previously implicated in telomerase action by a genetic screen. Mutation of the conserved reverse transcriptase amino acids led to telomere shortening and senescence in yeast, confirming the identity of this protein as a component of telomerase.

These papers [8•,9•] extended the family of telomerase reverse transcriptases (TERTs) to include members from human and fission yeast, demonstrating their conservation in a wide variety of organisms. Both papers also show that expression of the mRNA of human TERT correlates with telomerase activity in human cell lines and tissues, suggesting that a primary regulation of telomerase activity is provided by TERT expression.

Nat Genet 1997, 17:498-502. Telomerase activity was reconstituted in vitro by the expression of hTERT and telomerase RNA in rabbit reticulocyte lysates. Mutation of the conserved reverse transcriptase residues abolished this activity, providing direct evidence for the catalytic function of hTERT. Mutation of amino acids in the telomerase-specific motif 'T' also abolished activity, demonstrating that it is functionally important. This study suggests that TERT and the telomerase RNA form the minimal catalytic core of telomerase, although the contribution of other components of the reticulocyte lysate is not ruled out (see annotation [19•]).

Although the expression of hTERT and telomerase RNA in vitro are sufficient for telomerase activity, it is shown here that reconstitution is dependent on factors present in the rabbit reticulocyte lysate used in the transcription/translation reaction, specifically the chaperone proteins p23 and hsp90. They were shown to be associated with TERT by the yeast two-hybrid trap and by immunoprecipitation, and they co-immunoprecipitated with telomerase activity from human cells. An inhibitor of hsp90-p23 activity inhibited telomerase activity in cells, demonstrating that these proteins may also play a role in telomerase assembly in vivo.

This paper provides a mechanism by which the yeast telomere protein Rap1p may exert control over telomere length. Experiments in which a hybrid Rap1p-Gal4 protein was targeted to a Gal4 binding site next to the telomeric repeats of a yeast chromosome showed that the size of the telomeric tract subsequently decreased by an amount proportional to the number of Rap1p molecules targeted. Thus the number of Rap1p molecules provides a feedback mechanism for telomere length control.

Tankyrase, a novel protein that binds to TRF1, was identified through a two-hybrid screen. Tankyrase has a region homologous to PARP, (poly[adenosine diphosphate-ribose] polymerase), and like PARP has ribosylation activity. Evidence is presented that tankyrase ribosylates TRF1, at least in vitro, and thereby prevents it binding to telomeric DNA, providing a model for the regulation of TRF1 inhibition of telomerase.

This paper provides a function for the second human telomere-binding protein, TRF2. Expression of deletion mutants of TRF2 led to over 80% of the cells containing one or more end-to-end fused chromosomes, and evidence was presented that the fusions occurred by blunt-end ligation of telomeric DNA. Thus the protective role of telomeres is likely to be mediated by proteins such as TRF2.

Schizosaccharomyces pombe that lack TERT experience telomere shortening and senescence although, like Saccharomyces cerevisiae, a subpopulation of cells survives and divides indefinitely. This paper shows that these cells survive by one of two mechanisms: lengthening of telomeres by presumably recombinational means, like S. cerevisiae; or by circularising all three of their chromosomes. The DNA-binding protein Taz1p regulates the recombinational mode of telomere lengthening, as well as telomerase-mediated lengthening.

These two papers [47••,48••] provide a long-awaited demonstration that maintenance of telomeres can overcome senescence in at least some normal human cells. hTERT was ectopically expressed in BJ foreskin fibroblasts and RPE retinal epithelial cells, and all hTERT expressing clones had longer life spans in vitro than control cells. It is premature to conclude that these cells are immortalised but there is no doubt that they have an extended in vitro life span.

These cells have now achieved >200 PD in vitro, supporting the hypothesis that telomerase expression can immortalise human cells. The telomerase-expressing cells demonstrate normal cell-cycle checkpoint control, normal karyotypes and do not form tumours, making them valuable tools for the research of immortalisation.

Nature 1998, 396:84-88. This paper demonstrates that telomerase activity and maintenance of telomeres are not sufficient for the immortalisation of all human cell types. Although hTERT expression did achieve an extended life span for foreskin fibroblasts, immortalisation of mammary epithelial cells and foreskin keratinocytes also required the Rb/p16 growth control pathway to be inactivated. Thus, there are cell-type differences in the control of senescence.

This paper also demonstrates the extension of life span of normal human cells by hTERT expression. Interestingly, it is also shown that overexpression of the oncogene c-Myc induces expression of hTERT mRNA and telomerase activity in fibroblasts and epithelial cells. This provides a link between oncogenic transformation by c-Myc and telomerase activity; it is possible that the increased Myc activity in most tumours could be responsible for the high frequency of telomerase-expressing tumours.

Normal human uroepithelial cells were shown to lack telomerase activity, but activity was detected when these cells were cultured in vitro. Telomerase activity was also detected in bladder carcinomas. The authors interpret these results to indicate that telomerase activity is not related to malignant transformation. In addition, they find that uroepithelial cells in culture have telomerase activity and stable telomeres, yet still undergo senescence. This suggests that at least some cell types do not use telomere shortening as a primary signal for senescence.

This description of transgenic mice lacking the RNA component of telomerase has generated much uncertainty regarding the role of telomerase in cell growth and tumourigenesis. Telomerase-negative mice showed a progressive loss of telomeric DNA with each generation and an increase in chromosomal end fusions; however, cells from these mice showed no decrease in their ability to either immortalise in culture or form tumours in nude mice. Thus, either telomerase activation is not a prerequisite for continued cell growth or mice are different from humans in this regard.

The paper above [57••] generated much surprise that mice were able to reproduce for six generations in the absence of telomerase. This paper demonstrates that the progressive telomere shortening with successive generations does eventually affect cell viability, especially in highly proliferative organs such as the haematopoietic system. Spermatogenesis was defective in late-generation telomerase-negative animals, and no generation seven offspring were produced. This indicates that telomerase is indeed essential for the long-term viability of highly-proliferative cells and therefore of organisms.
