Mol Microbiol 1998, 29:111-124. PilQ proteins (also known as secretins) are required for type IV pilus biosynthesis in Neisseria meningitidis. In the 5′ domain of the open reading frame between four and seven repeats encoding PAKQQAA at the polypeptide level are present. The authors describe that the number of repeats affects pilus biosynthesis, not qualitatively but quantitatively. This for the first time suggested that repetitive DNA in prokaryotes might regulate the amount of protein synthesised.

The availability of this second whole genome sequence for a single bacterial species enabled for the first time a detailed comparison of the complete genomic repeat complement. In diverse gene classes clear polymorphism in SSRs is defined: in outer membrane protein genes (n = 5), flagella biosynthesis genes (n = 1), genes with regulatory functions (n = 1), restriction-modification systems (n = 4), gene homologues without a known function (n = 2) and in hypothetical open reading frames (n = 6) SSRs are now defined.

This is an excellent example of how whole genome sequences can be used for narrowing down SSRs and how the implications of repeat variability can be deduced from experimental data. The authors' conclusion that even in prokaryotes repeats are continuously maintained and generated by mutational pressure underlines the need for further research.
