This paper gives an overview of the recently discovered mevalonate-independent pathway of isoprenois biosynthesis, which is apparently localized to plastids in higher plants and algae. The author summarizes data from in vivo feeding experiments with stable isotopes and discusses the cooperation of both the mevalonate and thew mevalonate-independent pathway in isoprenoid metabolism.

The authors describe the cloning and heterologous expression of peppermint 1 -deoxyxylulose-5-phosphate synthase, a novel transketolase capable of catalyzing the proposed first reaction step in the mevalonate-independent pathway of isoprenoid biosynthesis. The synthase gene reveals an N-terminal signal sequence for plastidial targeting, consistent with the proposed subcellular localization of the pathway, and is highly expressed during the peak activity of monoterpene biosynthesis in peepermint essential oil glands.
