These studies demonstrate that two native yeast HAT complexes, NuA4 and NuA3, can be targeted to nucleosomes by a VP16 transcriptional activation domain to increase transcription in vitro. Significantly, a catalytic subunit of NuA4 is homologous to two proteins with demonstrated HAT activity, human Tip60 and yeast Esa1 p, and one putative HAT, human MOZ (see 'MOZ gene fusions' section and [54,58•,59•]).

The ADA and SAGA complexes contain the transcription-related HAT, Gcn5p, in association with adaptor proteins (Adas) and/or components of the basal transcription apparatus (Spts), thereby strengthening the link between histone acetylation and transcription activation.

Heterozygous mice with one inactivated allele of p300 display reduced viability during gestation but survivors suffer no gross phenotypic defects. Interestingly, mice heterozygous for both CBP and p300 also die during embryogenesis, suggesting that CBP and p300 share limited overlapping essential functions during development and that their combined gene dosage is important during development.

Proc Natl Acad Sci USA 1998, 95:4152-4157. The carboxy-terminal SET domain of MLL/ALL-1/HRX is shown to interact with hSNF5 by two-hybrid analysis, in vitro binding studies and co-immunoprecipitation from cultured cells or transgenic flies. This interaction suggests a mechanism for targeting SWI/SNF complexes to chromatin through a discrete functional domain of a non-histone chromatin component.

This study indicates a role for SWI/SNF in cancer in that somatic biallelic alterations of a human remodeling complex component were found in cells derived from malignant pediatric rhabdoid tumors. The alterations involve carboxy-terminal truncations of hSNF5/INI1, a region known to interact with hSNF2, an ATP-dependent catalytic subunit of the SWI/SNF complex.
