J Exp Med 1997, 185:367-371. Here, recombinant MHC class I molecules were refolded using a random peptide library. The bound peptides were subsequently identified in a pool-sequencing approach. The motif thus obtained, compared to that obtained from natural sources, differed at the carboxyl terminus - suggesting that molecules other than MHC class I were involved in the definition of simple natural motifs that are identified by pool sequencing.
