This paper reports the purification of a mammalian Arp2/3 complex, using actin cloud assembly around L. monocytogenes as an assay. The initial characterization of the mammalian Arp2/3 complex, its subunit stoichiometry and hydrodynamic properties are described.

Proc Natl Acad Sci USA 1998, 95:6181-6186. The Arp2/3 complex caps the pointed ends of actin filaments and nucleates the filaments that grow from their pointed ends. Using electron microscopy, the Arp2/3 complex was shown to be localized at filament ends and sides, and to form branching networks of actin filaments, in which a pointed end is attached to the side of another filament at a fixed angle of 70°.
