The clinical variability of HHG associated with AHC is exemplified by this case report of two patients. Measurement of circulating LH, follicle-stimulating hormone, and free glycoprotein hormone α-subunit together with a study of response to the administration of pulsatile GnRH indicate that HHG associated with AHC is caused by combined hypothalamic failure in GnRH release and pituitary detect in gonadotropin production.

Mol Endocrinol 1997, 11:1950-1960. A structural model of the DAX-1 protein C-terminus is presented on the basis of the similarity with apo-RXRα and holo-RARτ. Deletion analysis shows that the DAX-1 C-terminus contains a powerful cell type- and promoter-specific transcriptional silencing domain that needs two cooperating subdomains to function. One of these subdomains is located within helix H3, and the other is within helix H12. All mutations found in AHC kindreds impair transcriptional silencing by DAX-1.

WT1 missense mutations found in the Denys-Drash syndrome that produce mate pseudohermaphroditism impair transcriptional synergism with SF-1. DAX-1 is shown to antagonize the synergy between SF-1 and WT1 and to associate directly with SF-1 in the yeast two-hybrid system.

DAX-1 is shown to interact with SF-1 in a mammalian two-hybrid system. DAX-1 mutations R267P and, ΔV269, found in patients with AHC, significantly impair this interaction. Amino acids 226-230 and 437-447 of SF-1 are required for interaction with DAX-1. In addition, wild-type DAX-1 but not DAX-1 and ΔV269 DAX-1, can recruit the corepressor N-CoR to SF-1. The other corepressor, SMRT, cannot interact with DAX-1 in this assay. A point mutation in DAX-1 (F449D) is identified on the basis of sequence similarity with the orphan nuclear receptor Rev-Erb (which also interacts with N-CoR), which impairs its repression activity and interaction with N.CoR.

Stem-loop structures are present in both the dax-1 and StAR promoters, which are essential for transcriptional repression by DAX-1. Block of StAR gene expression by DAX-1 in stably transfected Y-1 cells blocks steroid synthesis.

These authors inactivated the Dax-1 gene in mice with the Cre-lox strategy, crossing cytomegalovirus-promoter Cre transgenic mice with mice carrying a IoxP-targeted allele. Male mice whose single DAX-1 allele was inactivated were apparently normal. The only anomaly in their adrenal cortex was failure in regression of the X zone after puberty. In addition, expression of pituitary LHβ, follicle-stimulating hormone-β, glycoprotein hormone α-subunit, and adrenocorticotropic hormone was normal, as were levels of circulating LH, follicle-stimulating hormone, and corticosterone. Testis weight decreased by approximately 50%. At 10 weeks of age, the seminiferous tubules showed clear signs of epithelial degeneration; at 14 weeks of age, complete loss of germ cells was observed. Leydig cell hyperplasia was present adjacent to degenerated tubules. Females that had both Dax-1 alleles inactivated were normally fertile and had only mild defects in follicle development.
