Is schizophrenia a lifetime disorder of brain plasticity, growth and aging?

DeLisi LE. Is schizophrenia a lifetime disorder of brain plasticity, growth and aging? Schizophr Res 1997; 23:119-129. This is an hypothesis about what the genes may be doing and how they connect with the known structural brain findings in schizophrenia.

Use of a quantitative trait to map a locus associated with severity of positive symptoms in familial schizophrenia to chromosome 6p
-
Brzustowicz LM, Honer WG, Chow EWC, Hogan J, Hodgkinson K, Bassett AS. Use of a quantitative trait to map a locus associated with severity of positive symptoms in familial schizophrenia to chromosome 6p. Am J Med Genet 1997; 61:1388-1396. This is a novel approach with a lot of potential. However, the results of this study should be interpreted with caution because the number of subjects and families is small, and the statistical approach needs verifying and confirmation in simulation studies for assurance about the frequency of false positives.

Reduced subcortical brain volumes in nonpsychotic siblings of schizophrenic patients: a pilot magnetic resonance imaging study. Am J Med Genet 1997; 74:507-514. This study, using a very small number of individuals, is preliminary and needs to be replicated in a larger sample, with other more advanced gray-white segmentation techniques and with a focus on families with more than one schizophrenic sibling so that the familial aspect to structural abnormalities, as well as whether they segregate with schizophrenia within families, can be determined.

NIMH Genetics initiative millennium schizophrenia consortium: Linkage analysis of African-American pedigrees
-
Kaufmann CA, Suarez B, Malaspina D, Pepple J, Svrakic D, Markel PD, Meyer J. NIMH Genetics initiative millennium schizophrenia consortium: linkage analysis of African-American pedigrees. Am J Med Genet 1998; 81:282-289. A very small number of families were used in these analyses. The cohort was split from the Faraone et al. [49] cohort based on their Afro-American heritage. There is no genetic evidence to date to do this.

Thome J, Foley P, Riederer P. Neurotrophic factors and the maldevelopmental hypothesis of schizophrenic psychoses. J Neural Transmission 1998; 105:85-100. This is a good review of relevant neurotrophic factors and how they may be related to the development of schizophrenia. Large CAG/CTG repeats are associated with childhood schizophrenia
-
Burgess CE, Lindblad K, Sidransky E, Yuan Q-P, Long RT, Breschel T, et al. Large CAG/CTG repeats are associated with childhood schizophrenia. Mol Psychiatry 1998; 3:321-327. According to the expanded repeat hypothesis, age of onset will be successively earlier from generation to generation. Thus, a study of children who already have developed schizophrenia would be an excellent focus for finding expanded repeats, it any patients with this diagnosis have them. This study, although small (n=36), found a significantly greater percentage of CAG/CTG repeats greater than 180 nucleotides in the patients (42%) compared with control individuals (29.5%). This difference was significant for male, but not female patients. The authors note, however, that there was an unusually low number of repeats in the control males and show preliminary data that this may be due to variable ethnic origin of groups.
