The authors describe the cloning of the p66Shc isoform, which differs from p52Shc/46Shc by an additional amino-terminal collagen homologous region (CH2). In contrast to p52/46Shc, p66Shc has an inhibitory effect on fos promoter activation upon EGF stimulation and the isolated CH2 domain retains this inhibitory effect.

Tyrosine phosphorylation of the EGF receptor by the kinase Jak2 is induced by growth hormone

The authors of this paper demonstrate tyrosine phosphorylation of the EGFR by Jak2 upon stimulation with growth hormone (GH). Ectopic expression of the EGFR or its kinase inactive mutant revealed that GH-induced MAP kinase activation and c-fos expression requires tyrosine phosphorylation of the EGFR but not its intrinsic kinase activity.

In this study, Smad 1 is shown to be a target of opposing regulatory input through RTKs and RS/TKs. Stimulation of epithelial lung cells with EGF induces a rapid increase in serine phosphorylation of Smad1 by the ERK family of MAP kinases. This phosphorylation in the linker region prevents nuclear accumulation of Smad1. In contrast, BMP-induced serine phosphorylation of Smad1 in the carboxy-terminal domain induces nuclear accumulation.
