Circ Res 1996; 79:840-848. This study documents the change in production of NO from the coronary circulation in conscious dogs during exercise. The production of NO across the heart was shown to increase during exercise, and was blocked by NLA (an NO inhibitor). After blockade of NO synthesis, increases in coronary blood flow were not affected, whereas oxygen consumption by the heart was increased at all levels of cardiac work. This suggests that NO plays a role in matching blood flow to tissue metabolism during exercise.

The role of NO on total oxygen consumption in conscious dogs was studied. This study found that total oxygen consumption was increased in conscious dogs after blockade of NO synthesis. This effect was not related to hemodynamic changes or changes in the activities of the central nervous system. The increased oxygen consumption was converted to heat.

This study shows that blockade of NO synthesis increases oxygen consumption in the kidney in conscious dogs, and renal efficiency for transporting sodium is decreased. Exogenous and endogenous NO also decreases oxygen consumption in the cortex and medulla of the kidney in vitro.

The changes in the production of NO across the heart and metabolic substrates during the development of pacing-induced heart failure, especially from compensation to decompensation state, were observed. This study found that the production of NO across the heart was significantly decreased during the decompensation state of heart failure. Most interestingly, during this state, the heart switched from fatty acid to glucose as the dominant fuel, suggesting that NO plays a role in the regulation of the metabolism of the heart.
